Kura Oncology develops small-molecule cancer drugs. Its first product, KOMZIFTI (ziftomenib), was approved by the FDA on November 13, 2025 for adults with relapsed or refractory acute myeloid leukemia (AML) carrying an NPM1 mutation, and product sales began that month. It competes directly with Syndax's REVUFORJ (revumenib), another approved menin inhibitor. Kura is also running two registrational Phase 3 trials of ziftomenib in newly diagnosed AML (the first, KOMET-017, is expected to report topline results in 2028), a trial in gastrointestinal stromal tumors with imatinib, and earlier programs including darlifarnib and KO-7246. The economics are shaped by a November 2024 agreement with Kyowa Kirin. Kyowa Kirin paid $330 million up front, and by the end of 2025 a further $240 million of development milestones had been achieved. Kura says it expects another $180 million of milestones in 2027-2028. In the United States Kura books KOMZIFTI sales and the two companies split commercial profits and losses 50/50. Outside the US, Kyowa Kirin sells the drug and owes Kura tiered double-digit royalties. Kyowa Kirin can end the agreement for convenience on 12 months' notice. What the business earns. Kura has never produced positive owner earnings. We measure owner earnings as operating cash flow with working-capital swings removed, less stock-based compensation and capital spending. On that basis the business consumed $27 million in 2016, $134 million in 2022, $186 million in 2024 and $292 million in 2025 ($3.33 per share). One accounting choice matters a lot. Most of the cash Kyowa Kirin has paid is recorded as deferred revenue, which Kura earns over time by doing development work. We did not count those payments as earnings in the year the cash arrived; they count only as the revenue is recognized. Without that adjustment 2024 would look cash-positive, which it was not in any recurring sense. In 2025 product sales were only $2.1 million (from the weeks after launch), and total revenue was $67.5 million, mostly collaboration revenue, against $251 million of R&D and $120 million of selling and administrative costs. The first half of 2026 ran at about $142 million of negative owner earnings, roughly $285 million a year. That is about the 2025 pace, not a repeat of the 57% widening from 2024 to 2025. We use the 2025 level as the starting point because it is the only full year since the Kyowa Kirin deal and the Phase 3 program began. Earlier years describe a smaller research company and would understate today's spending. The balance sheet. At June 30, 2026 Kura held $519 million of cash and short-term investments and owed $10 million on a term loan that matures in November 2027. Against that cash sits $454 million of deferred revenue: money Kyowa Kirin has already paid for work Kura still has to do. Shareholders' equity was only $48 million, about $0.54 per share. Kura says its end-2025 cash funds its plan into the fourth quarter of 2027, and that with the expected $180 million of milestones it can fund the program through the KOMET-017 results in 2028. The cases (discounted at 6%): Bear, $0.00. The milestones slip or Kyowa Kirin uses its termination right, and spending continues at the 2025-2026 pace of roughly $285-300 million a year. The cash runs out during 2027-2028, and the company would need new equity to continue. Without evidence that KOMZIFTI can cover the cost base, nothing is left for today's shares. Base, $0.13. The $180 million of milestones arrive in 2027 and 2028 as the contract and the company expect. Owner earnings stay at the demonstrated rate of about -$285 million a year through the end of 2028, when KOMET-017 reads out. By then the cash and the milestones are almost exactly used up. The small positive value comes only from the gap in timing between money in hand and money spent. We give no value beyond 2028 because KOMZIFTI does not yet have a sales history showing it can earn a profit. Bull, $0.54. The milestones arrive on schedule and the prepaid development work is completed within what Kyowa Kirin's payments cover. After 2028 the ziftomenib business runs at roughly break-even under the 50/50 US cost and profit share, which leaves shareholders with about today's book equity. What was left out and why. None of the cases credits growth in KOMZIFTI sales, milestones beyond the $180 million expected, approval in newly diagnosed AML, or value from darlifarnib or KO-7246. A launch without a sales record and trials that have not read out are not yet earnings the business has shown it can produce. These are the real sources of upside, and they are why the values above are so low compared with the cash on the balance sheet. What would make this wrong. If KOMZIFTI sales grow fast enough that the 50/50 US profit share turns clearly positive, or KOMET-017 succeeds and opens up newly diagnosed AML, the shares could be worth far more than these figures, and the estimate would be revised on that evidence. If spending rises further or the milestones do not arrive, the company will have to sell new shares, and existing holders will be diluted. The most recent quarterly report's narrative was not available to us. The June 30, 2026 balance sheet and first-half 2026 figures come from the financial data filed with that report, and the split of 2026 revenue between product sales and collaboration revenue could not be confirmed.
