Incyte is a profitable biotechnology company built on one molecule, ruxolitinib. Sold as Jakafi in the US ($3.09 billion in 2025), as the Opzelura cream ($679 million), and as Jakavi abroad by Novartis (which paid Incyte $458 million in royalties), that one drug produced roughly four-fifths of 2025 revenue of $5.14 billion. The rest comes from smaller cancer drugs (Iclusig, Pemazyre, Monjuvi, Zynyz), the 2025 launch of Niktimvo ($152 million in its first year), and royalties from Lilly's Olumiant. The company holds about $3.6 billion of cash and securities and has no borrowings beyond about $35 million of finance leases. The dated risk is patent expiry. The 10-K gives a minimum exclusivity date of 2028 for Jakafi, Jakavi and Opzelura; the composition-of-matter patents run to June 2028 and the phosphate-salt patents to December 2028 with pediatric extension. Six generic makers have challenged the Jakafi patents. Two (Hikma in October 2025 and Granules in February 2026) settled on undisclosed terms, and those cases are still pending. Opzelura has later formulation patents to 2031 and 2040, which four generic makers are also challenging, though Zydus has not challenged the 2031 patents. A once-daily Jakafi tablet awaits a regulatory decision expected in mid-2026, but it carries the same 2028 date, so it does not extend the cliff. US Medicare negotiation exemptions for Incyte's drugs run through 2028. Owner earnings swing widely because Incyte expenses large licensing and acquisition payments: $805 million to MorphoSys in 2020 and $679 million for Escient in-process research in 2024 produced negative years. Per share, owner earnings were $1.87 in 2021, $2.34 in 2022, $2.26 in 2023, -$0.62 in 2024 and $8.34 in 2025. The 2025 figure overstates the run rate. Removing working-capital movements adds back the $280 million cash settlement paid to Novartis against royalties accrued in prior years, a $242 million one-off settlement gain sits in profit, about $248 million of the year's tax expense was deferred rather than paid in cash, and $100 million of the milestone revenue was a one-time Lilly upfront. Adjusting for these, we put the current earning power at about $1.0 to 1.2 billion a year, or roughly $5 to $5.50 a share. We judge the level from 2021 to 2025 with those items stripped out rather than from any single year, and earlier years describe a smaller company. The settlement also gives Incyte a lasting benefit: it halved the royalty Incyte pays Novartis on US Jakafi sales from 2025, worth roughly $150 million a year until US exclusivity ends. Growth credited: owner earnings rose sharply in 2025, but that jump was mostly the one-off items above. The base case starts 2026 at about $1.05 billion, below the reported 2025 figure, and lets 2027 rise only modestly on Jakafi's demonstrated paid-demand growth, Opzelura's growth in the US and Europe, and Niktimvo's first full years. Nothing is credited for unapproved pipeline drugs (povorcitinib, the mutCALR antibody INCA033989) or for new acquisitions. These are real programs but not yet earnings, so they sit outside every case. Base case (median): Jakafi and Jakavi lose exclusivity in 2028 and follow the usual oral-small-molecule erosion, with US Jakafi revenue falling by roughly 80 to 90 percent within about two years. Opzelura holds part of its sales on its later cream patents. The non-ruxolitinib products (about $600 million of 2025 sales, growing) and Olumiant and Tabrecta royalties (exclusivity to 2032) are what remain. Against them sits an R&D budget of about $2 billion a year that management has shown no inclination to cut. Owner earnings run near $1.05 to $1.15 billion through 2027, about $0.9 billion in 2028, then fall to roughly $50 to 150 million a year from 2029 as the remaining products barely cover research spending. The base case credits no replacement blockbuster but does assume management trims spending enough to stay roughly breakeven, which is consistent with how Incyte behaved before Jakafi became large. The business is worth about $4.4 billion, plus about $3.1 billion of net cash (after keeping $300 million for operations and deducting finance leases and $121 million of acquisition-related contingent payments), or about $36 a share. Bear case (10th percentile): one or more of the four pending Jakafi challengers wins early, or the settled entrants launch before the end of 2028, so erosion starts during 2028. Incyte keeps spending at current levels on a pipeline that does not deliver on time, so losses run through 2029 to 2031. It also repeats its own record of an expensive deal that is later written off; it has done so with MorphoSys and Escient. The bear case charges one $1 billion acquisition of that kind, and the remaining franchise is worth nothing after 2031. The business is worth about $0.9 billion plus $3.1 billion of net cash, or about $19 a share. Most of the value in this case is the cash on the balance sheet and the earnings already near-certain before 2028. Bull case (90th percentile): every generic is held to the end of 2028, Opzelura keeps most of its sales on its 2031 patents, and Niktimvo, Zynyz, Monjuvi and the Olumiant royalties grow on their current trajectory. From 2029 the company earns about $450 million a year, fading to about $300 million as Olumiant and Tabrecta royalties lapse in 2032. It is worth about $8.5 billion plus net cash, or about $56 a share. Even the bull case gives no credit to pipeline drugs that are not yet approved. The previous analysis put the shares at $32, $57 and $105. These figures are lower mainly because we now judge that, once Jakafi loses exclusivity, the remaining products cannot carry the roughly $2 billion research budget without a replacement that has yet to be approved. We previously gave credit for povorcitinib and newer launches replacing much of the loss. Margin of safety price. We are fairly sure of earnings through 2028: Jakafi demand is documented, the patent dates are known, and the settlement with Novartis fixed the royalty cost. We are not sure what happens after 2028. That depends on unapproved drugs and on how much Incyte chooses to spend, both of which are outside our evidence. The worst plausible outcome is early generic entry combined with continued heavy research spending and another expensive acquisition write-off. The structure survives that, because there is no debt and the cash pile is large, but most of the operating value disappears. If we are wrong about the post-2028 business and it is worth nothing, the shares are still backed by about $15 a share of net cash plus roughly $14 to $15 a share of owner earnings over the next two and a quarter years, about $29 to $30 a share of hard value by the end of 2028, provided management does not spend it on a failed deal. From $18, that hard value alone implies about a 20 percent annual return over that period. Any lasting value in Opzelura, Niktimvo or the royalties is additional. The price assumes a holding period of about three years for the value to show, and during that time the company pays no dividend, so the return depends on the cash being kept or used well. That gives a margin of safety price of $18.

Sales geography
United States (all regions: Northeast, Midwest, West, South) for the large majority of product sales; Europe (France, Italy, Germany, Spain, United Kingdom) and Japan for a smaller share of product sales; worldwide royalties via Novartis (Jakavi, Tabrecta) and Eli Lilly (Olumiant).
Historical growth strategy
Builds on one in-house discovery (ruxolitinib) by extending it into new indications and formulations (Jakafi, Opzelura cream, once-daily XR), and spends heavily on in-licensing and acquisitions (MorphoSys tafasitamab, Escient, Syndax axatilimab partnership, Ariad European rights) to replace the franchise before patent expiry. Royalties from partners (Novartis, Lilly) add a second stream.
Chart analysis
A chartist would describe Incyte as a long-term range-bound stock: after a powerful run to a peak near $150 in 2015, the shares spent most of the following decade oscillating in a broad band of roughly $50 to $100, with repeated failures at the upper end and support in the mid-$50s. The pattern is one of a stock that has gone sideways for years while the business grew, with sharp gaps on clinical and legal news rather than steady trends. Rallies toward the top of the range have tended to stall, so a technician would want to see a decisive break and hold above the multi-year ceiling before calling a new uptrend, and would treat a break of the long-standing support as a sign that the patent-expiry concern is taking control.
