Entrada Therapeutics is a clinical-stage biotechnology company based in Boston. It has no product on the market. It is developing "endosomal escape vehicle" therapies for neuromuscular disease, including exon-skipping candidates for Duchenne muscular dystrophy (ENTR-601-44 is the most advanced, with ENTR-601-45 behind it) and a first ocular candidate, ENTR-801, announced in December 2025. Substantially all of its revenue to date has come from one agreement with Vertex for the myotonic dystrophy type 1 program VX-670. Entrada received a $223.7 million upfront payment plus a $26.3 million equity investment when the deal closed in February 2023, then milestone payments of $17.5 million (2023) and $75.0 million (2024). Revenue was $129.0 million in 2023, $210.8 million in 2024 and $25.4 million in 2025, because the upfront and milestone cash was recognised as revenue as the research work was done. Only $0.3 million of that deferred revenue remained at the end of 2025. How we measured earnings. We start from cash generated by operations and remove swings in working capital (including the large deferred-revenue movements from the Vertex deal), stock compensation and maintenance capital spending. On that basis the company's per-share owner earnings were about -$8.70 in 2021 (a year with an unrepresentative share count because the company listed in November), -$3.05 in 2022, -$0.33 in 2023, +$1.51 in 2024 and -$3.49 in 2025. The 2024 figure is the recognition of the Vertex upfront and milestone money, not a repeatable profit. The three years since the Vertex deal have a median of -$0.33 per share, which is not a usable earning power. Owner earnings fell from +$58.9 million in 2024 to -$144.4 million in 2025, and we assume no growth from that base. So we value the shares from the cash the company holds and what it will realistically spend, and we give no value to drug candidates that have not yet produced approved products. What the company holds and spends. At December 31, 2025 Entrada had $295.7 million of cash and marketable securities, and it says this funds operations into the third quarter of 2027. Operating cash use was $128.5 million in 2025 and $74.2 million in the first half of 2026 (about $12.4 million a month). By our estimate, cash and securities were therefore roughly $221 million at June 30, 2026 (a derived figure, since we did not have that quarter's balance sheet detail). Against that the company owes $48.5 million in lease payments on its Boston laboratory and office space (the balance at June 30, 2026), partly offset by sublease income of $2.1 million in 2025. Share count is about 42.3 million: 38.9 million shares outstanding plus 3.4 million pre-funded warrants issued in June 2024 that convert for a nominal price. Bear case, about -$0.16 per share. Entrada keeps spending at its current pace through its stated funding runway, into the third quarter of 2027, and its clinical results do not lead to any new payment. Fifteen months at about $12.4 million a month uses roughly $185 million of the estimated $221 million, leaving about $36 million against roughly $43 million of remaining lease obligations (the $48.5 million less about $5.6 million of lease principal paid over the period). On these numbers the company would owe slightly more than it holds, by about $7 million, and would have to raise new money at the expense of current holders. Base case, about $2.34 per share. Entrada has told investors it expects patient data from two ENTR-601-44 cohorts and one ENTR-601-45 cohort during 2026. We take those results as the point at which spending has to be justified. We do not credit value to programs that have not yet produced approved products, so continued spending after that point earns nothing in our valuation and we assume it stops at the end of 2026. Another six months at the current pace uses about $74 million, leaving about $147 million of cash; subtracting the $48.5 million of lease obligations leaves about $99 million, or about $2.34 per share. Bull case, about $4.01 per share. As in the base case, the company stops its own spending at the end of 2026. In addition, Vertex continues to advance VX-670, whose Phase 1/2 trial Vertex has said it expects to finish enrolling and dosing in mid-2026, and pays one more clinical-advancement milestone. We size it at $75.0 million, equal to the milestone paid in 2024, and expect it about a year from now, worth about $70.8 million today. The agreement allows for up to $485.0 million of further research, development, regulatory and commercial milestones, but only a milestone of this kind, with a payment already made in the past, is credited. That takes the value to about $170 million, or about $4.01 per share. No royalties or approved-product sales are credited. What we left out and why. We give no value to the Duchenne, ocular or myotonic dystrophy programs beyond the milestone above, because none has reached approval and there is no contract fixing their payments. We also do not assume that the company can raise new money on good terms. Cash use, rather than accounting profit, is the measure here, and it already includes interest earned on the company's cash. The company posted a net loss of $143.8 million in 2025 and has an accumulated deficit of $273.1 million. What could make these estimates wrong. Positive data from the 2026 Duchenne cohorts, or additional Vertex milestones beyond the single one we allow, would make the shares worth more than our bull case. Conversely, a bigger burn or an unsubleased lease would lower all three cases. This is a company whose value rests mainly on the science, which we cannot price from the filings, so the margin-of-safety price is set well below the base case.
