Business condition attached to the margin of safety The first pivotal Phase 3 trial of VAX-31 in adults (OPUS-1, results expected in the fourth quarter of 2026) shows it meets its non-inferiority goals against the pneumococcal vaccines now in use.
Vaxcyte is a clinical-stage vaccine developer with no products approved for sale and no product revenue. Its lead candidate, VAX-31, is a 31-strain pneumococcal conjugate vaccine for adults, aimed at the market served by Pfizer's Prevnar 20 and Merck's Capvaxive. The company has lost money every year, so there is no earnings history to value. Owner earnings (operating cash flow, less working-capital swings, stock compensation and capital spending) were negative every year from 2020 through 2025: roughly -$2.76, -$2.38, -$3.25, -$4.40, -$4.53 and -$6.01 per share. The loss grew as spending on trials and manufacturing rose. Cash used in operations was $655.6 million in 2025 and $513.1 million in the first half of 2026. Our reconciliation of net income to operating cash flow left a gap of about 6% to 8% in 2024 and 2025 that we could not fully tie to individual filing lines, and it does not change the conclusion. What we can value is the cash. At December 31, 2025 the company held $2,442.6 million of cash, cash equivalents and investments. Subtracting first-half 2026 operating cash use and capital spending gives roughly $1.92 billion, or about $13.30 per share on 144.5 million shares (the second-quarter 2026 count). This is an estimate, because the most recent quarterly balance sheet was not available to us in full. What is real and under way: the FDA granted VAX-31 Breakthrough Therapy designation for adults; the Phase 2 study compared it with Prevnar 20 in 1,015 adults aged 50 and older; and three Phase 3 trials started between December 2025 and February 2026 (OPUS-1, OPUS-2, OPUS-3). Results from OPUS-1 are expected in the fourth quarter of 2026 and from OPUS-2 and OPUS-3 in the first half of 2027, with a marketing application planned shortly after the last trial ends. What is not real yet: approval, a launch, pricing, or any sales. We do not credit growth that depends on those. Bear case, about $9.50 per share. The fourth-quarter 2026 results disappoint and the company stops the program. Spending continues at the first-half 2026 pace for the second half of 2026, leaving roughly $1.4 billion, or about $9.70 per share, and we shade that down for unquantified wind-down costs. Nothing is credited for products. Base case, about $13 per share. We credit only the cash on hand today and no future product sales. We do not deduct future trial spending either, because the Phase 3 program is already funded and under way with breakthrough designation, and we treat its cost and its worth as offsetting rather than assuming a gain or a loss. This is a judgement, not a forecast of what the company will earn. Bull case, about $13 per share. Even if the trials succeed on schedule, the vaccine has no approval, launch date or contracts today, so we credit no sales. The bull case therefore equals the base case. Real upside exists if VAX-31 is approved and captures sales, but we cannot point to evidence for its size and so leave it out. Margin-of-safety price, $6.00 per share. The company is spending about $1 billion a year. Continuing at that pace through the first-half 2027 trial results and the application would consume roughly $1.04 billion, leaving about $885 million, or about $6.10 per share. The margin-of-safety price is set at or below that level, so that it credits nothing for the vaccine program, and it applies only once the OPUS-1 results are in. What could make us wrong: a raise of new capital would add shares and dilute the per-share figures; spending could exceed the first-half pace; and a successful, approved vaccine would be worth far more than the cash we credit.

No product sales yet. The lead vaccine is being developed for approval by the U.S. Food and Drug Administration (Breakthrough Therapy designation granted); any first sales would be in the United States.
