Business condition attached to the margin of safety A clear, dose-dependent increase in frataxin levels from the RESTORE-FA Phase 1/2 trial (data expected second half of 2026) that shows the redesigned FA drug (DT-216P2) is working in patients.
Design Therapeutics is a clinical-stage biotechnology company applying its own small-molecule 'GeneTAC' platform to diseases caused by DNA repeat expansions. Its two active programs are DT-216P2 for Friedreich's ataxia (FA), currently in a Phase 1/2 dose-finding trial (RESTORE-FA) with a data update on the drug's effect on frataxin levels expected in the second half of 2026, and DT-168 for Fuchs endothelial corneal dystrophy (FECD), in an ongoing Phase 2 biomarker trial. Neither program has yet produced positive efficacy data, and the company has no approved products and no product revenue. As of June 30, 2026, the company held about $207 million in cash and short-term investments and carried no debt. It is spending roughly $55-65 million a year on research, development and overhead, funded from that cash balance and, periodically, from sales of stock under an existing at-the-market equity program (about $25 million was raised this way in 2025). Management has stated its cash is enough to fund operations for at least the next twelve months, but not enough on its own to carry either program through regulatory approval. Because neither program has produced positive clinical results yet, we have not assigned any value to the possibility that either drug eventually reaches the market - a hoped-for future is not evidence, and crediting it here would mean paying for a story rather than a fact. Instead, the value of the shares in all three cases comes almost entirely from the cash on the balance sheet, net of the spending we expect before any inflection point, discounted back to today. Bear case: the FA program's earlier drug formulation (DT-216P1) already fell short of its efficacy goal in 2023, which is why the company redesigned it into the current DT-216P2. If the redesigned drug again falls short in the 2026 readout, the company's own history suggests management is more likely to fund another attempt than to return cash to shareholders promptly. That persistence, funded from the same cash pile, consumes most or all of the balance and can leave existing shareholders with little to nothing once further, more dilutive financing becomes necessary. This case values the shares at roughly $(1.20). Base case: spending continues close to its current pace through the 2026 data and its aftermath, without either program producing a catalyst we can credit under our evidence standard, and gradually tapers as the company manages its remaining cash. This leaves the shares worth roughly their net cash value after expected spending - about $0.05. Bull case: both trials proceed on their currently disclosed timeline without a repeat of the regulatory delay the FA program experienced in 2025 (an FDA clinical hold from June to December 2025), and spending is managed more efficiently, preserving more of the cash for shareholders. Even here we have not assumed either drug is approved, since there is no positive efficacy data yet to support that. This case values the shares at roughly $0.68. What would move this range: a clean, dose-dependent readout from the FA trial in the second half of 2026 would be the first positive clinical signal either program has produced, and would justify revisiting the pipeline's value rather than treating it as a free option. A further regulatory setback, or a repeat of the pattern of funding another attempt after disappointing data, would push results toward the bear case. On accounting: we treated stock-based compensation as a real cost, subtracting it from operating cash flow since it dilutes shareholders even though it is non-cash on the income statement. The company's equipment spending is modest and, with no product revenue to grow, we treated all of it as maintenance-level capital. Given how wide this range is and how much of the downside case rests on spending the cash, the margin-of-safety price is set at a substantial discount to the company's cash value - around $0.20 a share. A materially higher margin-of-safety price could be justified if the FA trial's 2026 readout is positive and clean, which would be the first hard clinical evidence that the platform's science works in patients.
